  • [Title] Expression of Autophagy and Reactive Oxygen Species-Related Proteins in Lacrimal Gland Adenoid Cystic Carcinoma.
  • PURPOSE: To investigate the difference of expression of autophagy and reactive oxygen species (ROS) related proteins in adenoid cystic carcinoma (ACC) of lacrimal gland in comparison with ACC of salivary gland.
  • MATERIALS AND METHODS: Formalin-fixed, paraffin-embedded tissue samples from patients pathologically diagnosed as lacrimal gland ACC (n=11) and salivary gland ACC (n=64) were used.
  • The prognostic factors related to disease-free and overall survival (OS) in lacrimal gland ACC by log-rank tests, were determined.
  • RESULTS: GSTpi in stromal cells was more highly expressed in lacrimal gland ACC (p=0.006), however, MnSOD in epithelial cells was expressed more in salivary gland ACC (p=0.046).
  • CONCLUSION: This is the first study to demonstrate the expression of autophagy and ROS related proteins in lacrimal gland ACC in comparison with the salivary gland ACC, which would provide a basis for further study of autophagy and ROS mechanism as novel therapeutic targets in lacrimal gland ACC.

  • CONTEXT: The salivary glands are the site of origin of a wide variety of neoplasms and are also relatively uncommon.
  • The incidence of salivary gland tumors (SGTs) is claimed to be influenced by geographical and racial factors.
  • Histopathological diagnosis plays a major role in the diagnosis of these neoplasms, with very few contribution using special stains, immunohistochemistry and cytogenetic studies.
  • Pleomorphic adenoma (60.71%) and adenoid cystic carcinoma (14.94%) were the most common benign and malignant neoplasms.

  • There is a limited number of studies on the potential usage of this antibody in the diagnosis of salivary gland tumors on routine practice in cell blocks.
  • MATERIALS AND METHODS: Sixty-nine fine needle aspiration (FNA) cytologic materials of predominantly oncocytic morphology salivary gland tumors; acinic cell carcinoma (AciCC) (n = 8), adenoid cystic carcinoma (n = 2), pleomorphic adenoma (PA) (n = 22), Warthin tumor (WT) (n = 20), myoepithelioma (ME) (n = 5), benign oncocytoma (BeO) (n = 3), mucoepidermoid carcinoma (MEC) (n = 7), mammary analog salivary gland carcinoma (n = 2) were immunostained with DOG1 (clone K9) stain.
  • CONCLUSIONS: FNA is a common tool in the diagnosis and management of salivary gland tumors.
  • DOG1-K9 clone was very useful with a unique staining pattern of apical-luminal positivity in the differential diagnosis of AciCC from other oncocytic salivary gland tumors.

  • [Title] Immunohistochemical Expression of CD<sub>56</sub> and ALDH<sub>1</sub> in Common Salivary Gland Tumors.
  • Cancer stem cells, of which aldehyde dehydrogenase isoform 1 (ALDH<sub>1</sub>) is an immunohistochemical marker, are a group of tumorigenic cells which are involved in migration and tumor recurrences.
  • We aimed to evaluate the expression of ALDH<sub>1</sub> and CD<sub>56</sub> in common salivary gland tumors, as well as their relationship with each other and with a number of clinicopathologic factors.
  • MATERIALS AND METHODS: Forty-five paraffin blocks of salivary gland tumors (pleomorphic adenoma, mucoepidermoid carcinoma and adenoid cystic carcinoma, 15 samples each) were selected.
  • Malignant tumors were classified into two groups: low-grade (including mucoepidermoid carcinoma grade I) and high-grade (including mucoepidermoid carcinoma grade III and adenoid cystic carcinoma).
  • ALDH<sub>1</sub> overexpressed in all three salivary tumors, but not to statistically significant degree (P=0.54).
  • CONCLUSION: It appears that the number of NK cells and their function change in different types of salivary gland tumors (benign/malignant) and stroma.
  • NK cells are important components of the anti-tumor system; therefore immune dysfunction is associated with tumor progression in tumors of the salivary gland.
  • ALDH<sub>1</sub> overexpression suggests its role in tumorogenesis, but ALDH<sub>1</sub> is not involved in the morphogenesis of salivary gland tumors.
